چکیده

Action potential (AP) generation and propagation through the dendritic tree of pyramidal neurons has received much attention lately1. Experiments indicate that inhibitory postsynaptic potentials (IPSPs) have a decisive e ect in controlling action potential invasion in dendrites2, 3. However, these phenomena were observed in slices, and remain to be investigated under in vivo conditions where neurons are subject to intense synaptic activity. Here, we have combined computational models with intracellular recordings of neocortical pyramidal cells in order to investigate how spikes can be controlled by IPSPs in vivo.

Voltage- and site-dependent control of the somatic impact of dendritic IPSPs.

Inhibitory interneurons target specific subcellular compartments of cortical pyramidal neurons, where location-specific interactions of IPSPs with voltage-activated ion channels are likely to influence the inhibitory control of neuronal output. To investigate this, we simulated IPSPs as a conductance source at sites across the somato-apical dendritic axis (up to 750 microm) of neocortical layer...

The Journal of Neurophysiology Effect of Common Anesthetics on Dendritic Properties in Layer 5 Neocortical Pyramidal Neurons

Understanding the impact of active dendritic properties on network activity in vivo has so far been restricted to studies in anesthetized animals. However, to date no study has been made to determine the direct effect of the anesthetics themselves on dendritic properties. Here, we investigated the effects of three types of anesthetics commonly used for animal experiments (urethane, pentobarbita...

Spikelets in Pyramidal Neurons: Action Potentials Initiated in the Axon Initial Segment That Do Not Activate the Soma

Spikelets are small spike-like depolarizations that can be measured in somatic intracellular recordings. Their origin in pyramidal neurons remains controversial. To explain spikelet generation, we propose a novel single-cell mechanism: somato-dendritic input generates action potentials at the axon initial segment that may fail to activate the soma and manifest as somatic spikelets. Using mathem...

Postnatal development of dendritic synaptic integration in rat neocortical pyramidal neurons.

The dendritic tree of layer 5 (L5) pyramidal neurons spans the neocortical layers, allowing the integration of intra- and extracortical synaptic inputs. Here we investigate the postnatal development of the integrative properties of rat L5 pyramidal neurons using simultaneous whole cell recording from the soma and distal apical dendrite. In young (P9-10) neurons, apical dendritic excitatory syna...

Backpropagation of physiological spike trains in neocortical pyramidal neurons: implications for temporal coding in dendrites.

In vivo neocortical neurons fire apparently random trains of action potentials in response to sensory stimuli. Does this randomness represent a signal or noise around a mean firing rate? Here we use the timing of action potential trains recorded in vivo to explore the dendritic consequences of physiological patterns of action potential firing in neocortical pyramidal neurons in vitro. We find t...
